I'm trying to convert the txtsub (obtained from TitleLab exports
as Quicktime text) into .srt, but sub2srt keeps saying it can't
recognize the format... (I'm using the 0.5.3. version on OS X
10.3.7).

vobsub file created by mencoder
knorke $ file subtitles_de.*
subtitles_de.idx: ASCII text
subtitles_de.sub: MPEG sequence, v2, program multiplex
knorke $ sub2srt subtitles_de.sub
Could not detect subtitles_de.sub format!

How often do I need to say this?
If you have an .idx file then it's VOBSUB.
sub2srt does not and will not support this.

It's important what players & subtitlers support. And at least VSFilter (formerly known as (Direct)VobSub) DOES support <color>, <i> and <b> tags at least, if not other... (It's generally known that mplayer's subtitler sux.)
